This seminar introduces the newly installed Total Internal Reflection Fluorescence (TIRF) microscope in Alf Månsson’s Molecular Motors and Bionano Laboratory, which is available for use by researchers at KOB. The talk will provide a brief overview of the fundamental principles of TIRF microscopy, the instrumental setup, followed by highlighting the key features of the microscope, and a general overview of its applications.
The seminar will also demonstrate how Total Internal Reflection Fluorescence (TIRF) microscopy is used for real-time observation of actin–myosin interactions at the single-molecule level in the presence of fluorescently labelled ATP. Using this technique, we also study how mutations associated with hypertrophic cardiomyopathy (HCM) alter myosin kinetics and assess the modulatory effects of cardiac myosin–targeting drugs, including Omecamtiv Mecarbil and Mavacamten.
